Lineage-level species distribution model to assess the impact of climate change on the habitat suitability of Boleophthalmus pectinirostris

Global climate change has profound impacts on the habitats of marine organisms, and predicting the habitat changes of species under climate change conditions is crucial for species sustainability. Boleophthalmus pectinirostris is an intertidal fish species that holds significant ecological and economic value. To better protect and manage its resources, this study aimed to predict its current potential distribution and habitat changes under different climate scenarios in the future. This study firstly quantified the hypervolume niches of the three lineages (AE1, AE2, and AES lineages) and compared the niche differentiation among them. Furthermore, this study constructed species-level and lineage-level species distribution models (SDMs) to assess the impact of climate change on the habitat suitability of B. pectinirostris. The result of the niche differentiation assessment showed that there was marked differentiation in niches among the three lineages. The responses of different lineages to environmental variables were different, suggesting that lineage-level models may provide more accurate prediction results. According to the model predictions, the AES may have greater resilience to climate change and may experience habitat expansion in the future, while the AE1 and the AE2 may face habitat loss in some regions. Climate change-driven shifts in oceanic conditions were anticipated to affect the distribution and community structure of marine organisms. This study assessed the impact of climate change on the suitable habitat range of three lineages of B. pectinirostris using SDMs. Consistent with previous studies, the results of our study indicated that lineage-level SDMs may be more reliable than species-level SDMs for species with population differentiation in terms of the accuracy of predictions. In addition, considering the vulnerability of the AE1 and AE2 lineages to climate change, conserving these two lineages should be given a higher priority. The results of this study will provide important information for the future management and conservation of this species.

Cryptic diversity of mudskipper genus Boleophthalmus (Gobiiformes: Oxudercidae) from the north coast of East Java, Indonesia

Biodiversitas 25: 412-420. Mudskippers of genus Boleophthalmus are commonly found in the Indo-Pacific area. This study aimed to examine the diversity of the genus Boleophthalmus from the north coast of East Java Province, Indonesia by employing the mitochondrial COI gene for DNA barcoding and morphological characterization. Ten individuals of this genus were analyzed. According to the similarity percentage evaluated using Basic Local Alignment Search Tool (BLAST) and Barcode of Life Data (BOLD) identification engines, all the individuals were identified as Boleophthalmus boddarti Pallas, 1770. However, phylogenetic analysis revealed that three of the individuals were identified as B. boddarti, and seven were identified as Boleophthalmus pectinirostris Linnaeus, 1758 originated from East Java, which is separated from the clade of B. pectinirostris from East Asia population. These results were supported by the genetic distance of 8.93%-13.81% and the morphological characteristics determined by principal component analysis (PCA). PCA revealed that B. boddarti and B. pectinirostris are primarily distinguished by the number of (longitudinal) scales (69-75 versus 113-123), length of second dorsal fin base (42.3-43.9 versus 38.1-49.0), and caudal fin length (18.1-22.4 versus 17.5-21.6). Further detailed studies are needed to clarify whether the seven individuals belonging to B. pectinirostris complex should be regarded as a new species or a new subspecies.

Molecular characterization and functional study of a galectin-9 from a teleost fish, Boleophthalmus pectinirostris

Galectin-9, a tandem-repeat galectin, plays an important role in the regulation of innate immune response against various microbial infections. Here, galectin-9 from mudskipper (Boleophthalmus pectinirostris) was identified and named as BpGal-9. Putative BpGal-9 contains two conserved carbohydrate recognition domains (CRDs), one CRD within N-terminal (N-CRD) and the other one within C-terminal (C-CRD). Multi-alignment analysis indicated that BpGal-9 shared the highest amino acid sequence identity of 64.3 % with that of Southern platyfish (Xiphophorus maculatus). Phylogenetic analysis showed that BpGal-9 grouped tightly with other teleosts galectin-9 and was most closely related to that of Southern platyfish. BpGal-9 transcripts were more abundant in the intestine, and its expression upregulated significantly in the intestine, kidney, spleen, gills, and skin after Edwardsiella tarda infection. Meanwhile, BpGal-9 expression significantly increased in hemocytes and serum of mudskipper infected by E. tarda. The recombinant BpGal-9 (rBpGal-9) and rBpGal-9C-CRD could agglutinate all tested bacteria, whereas rBpGal-9N-CRD could only agglutinate three kinds of bacteria. When targeting the same bacteria, rBpGal-9 showed stronger agglutinating activities than rBpGal-9C-CRD or rBpGal-9N-CRD. In addition, the induction effect of three recombinant proteins on the mRNA expression of anti-inflammatory cytokines (BpIL-10 and BpTGF-β) was better than that on the pro-inflammatory cytokines (BpIL-1β and BpTNF-α). Our result suggested that the N-CRD and C-CRD of galectin-9 contribute differently to its multiple functions in innate immunity in teleosts.

Genomics comparisons of three chromosome-level mudskipper genome assemblies reveal molecular clues for water-to-land evolution and adaptation

IntroductionMudskippers are a large group of amphibious fishes that have developed many morphological and physiological capacities to live on land. Genomics comparisons of chromosome-level genome assemblies of three representative mudskippers, Boleophthalmus pectinirostris (BP), Periophthalmus magnuspinnatus (PM) and P. modestus (PMO), may be able to provide novel insights into the water-to-land evolution and adaptation. MethodsTwo chromosome-level genome assemblies for BP and PM were respectively sequenced by an integration of PacBio, Nanopore and Hi-C sequencing. A series of standard assembly and annotation pipelines were subsequently performed for both mudskippers. We also re-annotated the PMO genome, downloaded from NCBI, to obtain a redundancy-reduced annotation. Three-way comparative analyses of the three mudskipper genomes in a large scale were carried out to discover detailed genomic differences, such as different gene sizes, and potential chromosomal fission and fusion events. Comparisons of several representative gene families among the three amphibious mudskippers and some other teleosts were also performed to find some molecular clues for terrestrial adaptation. ResultsWe obtained two high-quality haplotype genome assemblies with 23 and 25 chromosomes for BP and PM respectively. We also found two specific chromosome fission events in PM. Ancestor chromosome analysis has discovered a common fusion event in mudskipper ancestor. This fusion was then retained in all the three mudskipper species. A loss of some SCPP (secretory calcium-binding phosphoprotein) genes were identified in the three mudskipper genomes, which could lead to reduction of scales for a part-time terrestrial residence. The loss of aanat1a gene, encoding an important enzyme (arylalkylamine N-acetyltransferase 1a, AANAT1a) for dopamine metabolism and melatonin biosynthesis, was confirmed in PM but not in PMO (as previously reported existence in BP), suggesting a better air vision of PM than both PMO and BP. Such a tiny variation within the genus Periophthalmus exemplifies to prove a step-by-step evolution for the mudskippers’ water-to-land adaptation. ConclusionThese high-quality mudskipper genome assemblies will become valuable genetic resources for in-depth discovery of genomic evolution for the terrestrial adaptation of amphibious fishes.

A progestin regulates the prostaglandin pathway in the neuroendocrine system in female mudskipper Boleophthalmus pectinirostris

Sex hormones regulate the reproductive cycle through brain-pituitary axis, but the molecular mechanism is still enigmatic. In the reproductive season, the mudskipper Boleophthalmus pectinirostris possesses a semilunar periodicity spawning rhythm, which coincides with the semilunar periodicity variations in 17α-hydroxyprogesterone, the precursor of 17α,20β-dihydroxy-4-pregnen-3-one (DHP), a sexual progestin in teleosts. In the present study, we investigated in vitro the brain transcriptional differences between DHP-treated tissues and control groups using RNA-seq. Differential expression analysis revealed that 2700 genes significantly differentially expressed, including 1532 up-regulated and 1168 down-regulated genes. The majority of prostaglandin pathway-related genes were dramatically up-regulated, especially the prostaglandin receptor 6 (ptger6). Tissue distribution analysis revealed that ptger6 gene was ubiquitously expressed. In situ hybridization results showed that ptger6, nuclear progestin receptor (pgr), and DHP-induced c-fos mRNA were co-expressed in the ventral telencephalic area, the ventral nucleus of ventral telencephalic area, the anterior part of parvocellular preoptic nucleus, the magnocellular part of magnocellular preoptic nucleus, the ventral zone of periventricular hypothalamus, the anterior tubercular nucleus, the periventricular nucleus of posterior tuberculum, and the torus longitudinalis. DHP significantly enhanced promoter activities of ptger6 via Pgr. Together, this study suggested that DHP regulates the prostaglandin pathway in the neuroendocrine system of teleost fish.

Cytoarchitecture of Mudskipper (Boleophthalmus pectinirostris) Brain

Mudskippers are intertidal burrowing fish with unique living habits. So far, studies on the cytoarchitecture of the brain in fish with such behaviors remain limited. Therefore, documenting the neuroanatomy of this animal is of interest because of its unique characteristics. In this study, we examined the cytoarchitecture of mudskipper (Boleophthalmus pectinirostris) brain and investigated whether it has any peculiarities in its brain structures. In general, the basic composition, morphology, and organization of mudskipper brain do not vary markedly from those of other teleosts. The main differences appear in the telencephalon and diencephalon. In addition to Nissl staining, immunostainings for catecholaminergic and cholinergic systems were performed to help identify certain nuclei. The results showed that the number of subdivisions of the central division of pallium, lateral division of pallium, and medial division of pallium were different with other teleost species. In addition, some diencephalic nuclei, including the nucleus subglomerulosus, lateral thalamic nucleus, and intermediate superficial pretectal nucleus, were absent, which suggests the corresponding functions, such as visual or gustatory function, are less developed or specialized in B. pectinirostris. These results will provide a fundamental neuroanatomical basis for future studies on neuroendocrine regulation of behavior in intertidal burrowing fish.

Accumulation of nutrients and potentially toxic elements in plants and fishes in restored mangrove ecosystems in South China.

Mangroves are highly dynamic ecosystems that offer important services such as maintaining biodiversity, filtering pollutants, and providing habitats for fishes. We investigated the uptake and accumulation of nutrients and potentially toxic elements in mangrove plants and fish to better understand the role of mangrove restoration in maintaining mangrove biota quality. In mangrove plants, the average bioconcentration factors of nutrients and potentially toxic elements were in the order P > Pb > Mn > Mg > Se > Zn > Hg > Cu > Cd > As > Co > Cr > Ni > Fe > V > Sb, where only P (all plant species) and Pb (Sonneratia apetala Buchanan-Hamilton) had a BCF > 1.0 in mangrove plants. In general, Sonneratia spp. had better performances than Kandelia candel (Linn.) Druce, Aegiceras corniculatum (Linn.) Blanco and Acanthus ilicifolius L. Sp. in terms of nutrient uptake and toxic metal(loid)s accumulation, and the best uptake capacity was found in S. apetala. Fast growth and easy adaptation make S. apetala suitable for a restored mangrove ecosystem, but continual management is needed to prevent its suppression of mangrove species diversity. The concentration of As, Cd, Hg, Cu, Cr and Pb in the mangrove sediment were 30-220% higher than the Chinese National Standard of Marine Sediment Quality Class I limits, suggesting that the sediments were unsuitable for aquaculture and nature reserves. Although a higher toxic metal(loid)s concentration in the sediment was found, the target hazard quotient (THQ) of this toxic metal(loid)s in 5 mangrove habitat fishes was <1.0, except THQ of Pb in Boleophthalmus pectinirostris Linnaeus was 1.17, and THQ of Cr in Bostrychus sinensis Lacépède was 1.12. The low THQ (less than 1.0) of mangrove habitat fishes suggested that the restored mangrove system could alleviate the bioaccumulation of toxic metal(loid)s in mangrove fish.

The gastric proton pump in gobiid and mudskipper fishes. Evidence of stomach loss?

Stomach loss has occurred independently multiple times during gnathostome evolution with notable frequency within the Teleostei. Significantly, this loss of acid-peptic digestion has been found to correlate with the secondary genomic loss of the gastric proton pump subunits (atp4a, atp4b) and pepsinogens/pepsins (pga, pgc). Gastric glands produce gastric juice containing the acid and pepsin and thus their presence is a hallmark feature of a digestive system capable of acid-peptic digestion. However, in gobiid fishes although oesogaster and gastric glands have been identified histologically, their functional significance has been questioned. In the present study we address whether the gastric proton pump is present and expressed in gastric glands of the goby Neogobius species (Gobiidae) and in members of the family Oxudercidae, a group of amphibious gobiid fishes commonly known as mudskippers (genera: Periophthalmus, Boleophthalmus, Periophthalmodon and Scartelaos). We confirmed the presence of gastric glands and have immunohistochemically localized gastric proton pump expression to these glands in Neogobius fluviatilis and Periophthalmus novemradiatus, Periophthalmus barbarus and Boleophthalmus boddarti. Genome analysis in Neogobius melanostomus, Periophthalmus magnuspinnatus, Scartelaos histophorus, Boleophthalmus pectinirostris, and Periophthalmodon schlosseri revealed the presence of both atp4a and atp4b subunit orthologues in all species in a conserved genomic loci organization. Moreover, it was possible to deduce that the complete open reading frame and the key functional amino acid residues are present. The conserved expression of the gastric proton pump provides clear evidence of the potential for gastric acid secretion indicating that acid digestion is retained in these gobiid fishes and not lost.

A short peptidoglycan recognition protein protects Boleophthalmus pectinirostris against bacterial infection via inhibiting bacterial activity

Peptidoglycan recognition proteins (PGRPs) belong to a member of pattern-recognition receptors (PRRs), which proposed as antibacterial protein. The present study investigated the antibacterial effect of BpPGRP5 in great blue-spotted mudskipper (Boleophthalmus pectinirostris). BpPGRP5 transcript was detected in all tested tissues with the highest expression level in spleen, and its expression was significantly upregulated in spleen, intestine, and kidney following Aeromonas veronii infection. rBpPGRP5 was found to interact with several polysaccharides and bacteria, including Gram-negative bacteria (Escherichia coli and A. veronii) and Gram-positive bacteria (Listeria monocytogenes and Staphylococcus aureus). rBpPGRP5 inhibited the proliferation of E. coli, S. aureus, L. monocytogenes, and A. veronii in a Zn2+-dependent manner. Furthermore, in vivo studies revealed that intraperitoneal injection of rBpPGRP5 improved the survival rate of A. veronii-infected B. pectinirostris, accompanied by decreased bacterial load in the blood, kidney, intestine, and spleen. Taken together, our results indicated that BpPGRP5 is an antimicrobial protein that protects B. pectinirostris against bacterial infection.

Chromosome-Level Genome Assembly of Acanthogobius ommaturus Provides Insights Into Evolution and Lipid Metabolism

Acanthogobius ommaturus is a large, fast-growing annual fish widely distributed in coastal and estuarine areas. The adults will die after breeding, and its life cycle is only 1 year. The first chromosome-level genome assembly of A. ommaturus was obtained by PacBio and Hi-C sequencing in this study. The final genome assembly after Hi-C correction was 921.49 Mb, with contig N50 and scaffold N50 values of 15.70 Mb and 40.99 Mb, respectively. The assembled sequences were anchored to 22 chromosomes by using Hi-C data. A total of 18,752 protein-coding genes were predicted, 97.90% of which were successfully annotated. Benchmarking Universal Single-Copy Orthologs (BUSCO) assessment results for genome and gene annotations were 93.6% and 84.6%, respectively. A. ommaturus is phylogenetically closely related to Periophthalmodon magnuspinnatus and Boleophthalmus pectinirostris, diverging approximately 31.9 MYA with the two goby species. The A. ommaturus genome displayed 597 expanded and 3,094 contracted gene families compared with the common ancestor. A total of 1,155 positive selected genes (PSGs) (p &amp;lt; 0.05) were identified. Based on comparative genomic analyses, we obtained several expanded genes such as acsbg2, lrp1, lrp6, and znf638 involved in lipid metabolism. A total of twenty candidate genes were identified under positive selection, which associated with lifespan including ercc6, igf1, polg, and tert. Interspecific collinearity analysis showed a high genomic synteny between A. ommaturus and P. magnuspinnatus. The effective population size of A. ommaturus decreased drastically during 200–100 Ka because of Guxiang ice age and then increased gradually following warm periods. This study provides pivotal genetic resources for in-depth biological and evolutionary studies, and underlies the molecular basis for lipid metabolism.

Pandemic Covid-19 require for the implementation of restrictions on social interaction and outdoor activities. A decrease in the activity if it lasts a long time will have an impact on the decline in the income of the community. This is an effect of the purchasing power of the basic needs as well as the fulfillment of nutrition. While the conditions of the pandemic requires the fulfillment of nutrition, especially protein as a nutrient in maintaining the durability of the body. Mudskipper (Boleophthalmus pectinirostris) can be a cheap source of protein and easily obtainable by the people who live in the coastal region. This study aims to introduce the coastal communities in the Village Sriminosari, East Lampung Regency against potential mudskipper as a source of protein that are easily obtained during a pandemic without having to conduct operations from the sea, and strategies to increase its economic value with processed into fish nuggets. Research methods include field survey, observation or direct observation, discussion and interviews using research instruments in the form of questionnaires. Respondents were selected using the method of withdrawal of random samples of simple (simple random sampling). Testing of raw materials mudskipper using sheets of organoleptic test raw material in accordance with SNI 2729-2013. Test sensory processed nuggets using sheet test sensory tools reference SNI 2346:2015. The results of the research show the value of organoleptic mudskipper as a raw material that is 7,59 ? µ ? 7,73, and the value of the test sensory mudskipper nugget amounted to about 7.42? ? ? 7,74. This study provides knowledge to the community that mudskipper can be utilized in the fulfillment of nutrition, especially protein, answer the public stigma that mudskipper non-toxic and can be consumed as well as pose a public initiative to develop nuggets but by utilizing the raw material of other types of fish are abundant in the current season, but have no economic value in terms of price.

A high-resolution genome of an euryhaline and eurythermal rhinogoby (Rhinogobius similis Gill 1895).

Rhinogobius similis is distributed in East and Southeast Asia. It is an amphidromous species found mostly in freshwater and sometimes brackish waters. We have obtained a high-resolution assembly of the R. similis genome using nanopore sequencing, high-throughput chromosome conformation capture (Hi-C), and transcriptomic data. The assembled genome was 890.10 Mb in size and 40.15% in GC content. Including 1373 contigs with contig N50 is 1.54 Mb, and scaffold N50 is 41.51 Mb. All of the 1373 contigs were anchored on 22 pairs of chromosomes. The BUSCO evaluation score was 93.02% indicating high quality of genome assembly. The repeat sequences accounted for 34.92% of the whole genome, with retroelements (30.13%), DNA transposons (1.64%), simple repeats (2.34%), and so forth. A total of 31,089 protein-coding genes were predicted in the genome and functionally annotated using Maker, of those genes, 26,893 (86.50%) were found in InterProScan5. There were 1910 gene families expanded in R. similis, 1171 gene families contracted and 170 gene families rapidly evolving. We have compared one rapidly change gene family (PF05970) commonly found in four species (Boleophthalmus pectinirostris, Neogobius melanostomus, Periophthalmus magnuspinnatus, and R. similis), which was found probably related to the lifespan of those species. During 400–10 Ka, the period of the Guxiang Ice Age, the population of R. similis decreased drastically, and then increased gradually following the last interglacial period. A high-resolution genome of R. similis should be useful to study taxonomy, biogeography, comparative genomics, and adaptive evolution of the most speciose freshwater goby genus, Rhinogobius.

Expressions of melanopsins in telencephalon imply their function in synchronizing semilunar spawning rhythm in the mudskipper Boleophthalmus pectinirostris

The mudskipper Boleophthalmus pectinirostris inhabits intertidal mudflats, exhibiting semilunar reproductive rhythms. To investigate whether melanopsin is possibly involved in the synchronization of the semilunar spawning rhythm in the female mudskipper, we first cloned all four melanopsin subtypes (opn4m1, opn4m3, opn4x1, opn4x2) in B. pectinirostris. Results from RTq-PCR showed that significantly higher transcription levels of all four melanopsin subtypes were observed in the eyes rather than other tissues. In brain, all four melanopsin subtypes were also detectable in different regions, including the telencephalon, in which the expression of melanopsin has not been reported in other teleosts. The transcription levels of opn4m3 and opn4x1 in the telencephalon exhibited a daily fluctuation pattern. When females entered the spawning season, opn4m1 and opn4x1 transcript levels increased significantly in the telencephalon. During the spawning season, the transcript levels of opn4m3 and opn4x1 in the telencephalon appeared to have a cyclic pattern associated with semilunar periodicity, exhibiting two cycles with a peak around the first or the last lunar quarters. Results from ISH showed that, opn4x1 mRNA was localized in the medial of dorsal telencephalic area, dorsal nucleus of ventral telencephalic area (Vd), ventral nucleus of ventral telencephalic area (Vv), anterior part of parvocellular preoptic nucleus, magnocellular part of the magnocellular preoptic nucleus (PMmc), habenular and ventral zone of hypothalamus. Intriguingly, gnrh3 mRNA was also located in Vd, Vv and PMmc. Taken together, our results suggested that melanopsins, e.g. opn4x1, expressed in the telencephalon might mediate semilunar spawning activity in the female mudskipper.

Evaluation of the acute toxic effects of crude oil on intertidal mudskipper (Boleophthalmus pectinirostris) based on antioxidant enzyme activity and the integrated biomarker response

With the development of marine oil industry, oil spill accidents will inevitably occur, further polluting the intertidal zone and causing biological poisoning. The muddy intertidal zone and Boleophthalmus pectinirostris were selected as the research objects to conduct indoor acute exposure experiments within 48 h of crude oil pollution. Statistical analysis was used to reveal the activity changes of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST) in the gills and liver of mudskipper. Then, integrated biomarker response (IBR) indicators were established to comprehensively evaluate the biological toxicity. The results showed that the activities of SOD, CAT and GST in livers were higher than those in gills, and the maximum induction multipliers of SOD, CAT and GPx in livers appeared earlier than those in gills. Both SOD and GPx activities were induced at low pollutant concentrations and inhibited at high pollutant concentrations. For the dose-effect, the change trends of CAT and SOD were roughly inversed. There was substrate competition between GPx and CAT, with opposite trends over time. The activating mechanism of GST was similar to that of GPx, and the activation time was earlier than that of GPx. In terms of dose-effect trends, the IBR showed that the antioxidant enzymes activities in biological tissues were induced by low and inhibited by high pollutant concentrations. Overall, SOD and GPx in gills and CAT and GST in livers of the mudskippers were suitable as representative markers to comprehensively analyze and evaluate the biotoxicity effects of oil pollution in the intertidal zone. The star plots and IBR values obtained after data standardization were consistent with the enzyme activity differences, which can be used as valid supplementary indexes for biotoxicity evaluation. These research findings provide theoretical support for early indicators of biological toxicity after crude oil pollution in intertidal zones.

Morphology of the respiratory vasculature of the mudskipper Boleophthalmus pectinirostris (Gobiidae: Oxudercinae).

The gross morphology of the circulatory system in the amphibious mudskipper, Boleophthalmus pectinirostris, conforms with the typical teleost configuration, in which gills and systemic vascular beds are connected in series. However, at the microscopic level, the vasculatures of the respiratory organs, the inner epithelium of the bucco-opercular cavity, gills and skin, all show specializations for aerial gas exchange. The epithelium of the bucco-opercular cavity is heavily vascularized by respiratory capillaries that are derived from systemic arteries of the head, mainly branches of the hyomandibular artery and the dorsal opercular artery. The respiratory circuit of the secondary lamellae of the gills consists of 15-17 channels running in parallel, unlike the lacuna-like blood space of aquatic fishes. The most notable specialization is found in the microcirculation of the respiratory papillae in the skin. Each respiratory papilla is supplied by an arteriole that is derived from a systemic artery, mainly the cranial artery in the head and the segmental artery in the trunk. The arteriole divides several times along its course to the apical region of a papilla, where the branches split into approximately 65 capillaries that radiate to the periphery of the papilla. The capillaries twist 5-10 times before they unite to form the venules that encircle maximally half the circumference of a papilla. A variable number of venules merge into a vein, which progressively coalesces with veins from other papillae. There is no morphological specialization that separates oxygen-rich effluent blood of the epithelia of the bucco-opercular cavity and the respiratory papillae of the skin from the oxygen-poor systemic venous blood. The ecophysiological implications of these findings are discussed in relation to the environmental conditions that B. pectinirostris experience during tidal cycles in the warm months and during overwintering.

Effects of underwater and semi-aquatic environments on gut tissue and microbiota of the mudskipper Boleophthalmus pectinirostris.

In both underwater and semi-aquatic environments, the gut microbiota is of particular physiological importance for amphibious animals, given that the gut tract is among those organs in direct communication with the external environment. In this study, we examined the effects of these contrasting environments on the dominant bacteria in the guts of the amphibious mudskipper Boleophthalmus pectinirostris. Compared with the guts of normal mudskippers, in which the dominant bacteria were identified as Vibrio and Faecalibacterium, we found that Acinetobacter, Shigella, and Bacillus predominated in their guts after exposure to the semi-aquatic environment, whereas Escherichia, Bacteroides, and Bacillus were more prevalent in the guts in the underwater environment. The total number of cultured gut bacteria decreased significantly in the semi-aquatic environment. In semi-aquatic mudskippers, we also detected reductions and increases in the length and width of gut villi, respectively, whereas the width of gut villi declined and the number of goblet cells increased significantly in mudskippers maintained underwater. The mRNA expression of multiple gut transporters for glucose, long-chain fatty acids, and amino acids was found to increase markedly in both underwater and semi-aquatic environments, with the expression of most transporters being significantly higher in those mudskippers exposed to an underwater environment. Furthermore, we detected significant increases in the mRNA expression of pro-inflammatory cytokine transcripts in the guts of both underwater and semi-aquatic mudskippers on days 2, 4, and 6 of exposure, whereas the expression of IL-10 and TGFβ mRNA was more pronounced on days 4 and 8, respectively. Comparatively, we found that expression levels of cytokines in the guts of underwater mudskipper were substantially higher than those in the guts of semi-aquatic mudskippers. Collectively, our findings revealed notable differences in the gut microbiota and energy metabolism requirements of mudskippers exposed to underwater and semi-aquatic conditions, thereby providing a theoretical basis explaining the maintenance of a homeostatic state in mudskippers that constantly transition between these contrasting amphibious habitats.

The complete mitochondrial genome of the mudskipper, Boleophthalmus pectinirostris (Gobiiformes, Oxudercidae) from Beibu Bay

The mudskipper, Boleophthalmus pectinirostris (B. pectinirostris), is an amphibious fish that lives in the intertidal mudflats. It is a cultured economic fish with nutritional and pharmacological value. Here, we report the complete mitochondrial genome sequence of B. pectinirostris, which is 17,111 base pairs (55.3% A + T content) in length and consists of 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs, and a 1453 bp D-loop region. The complete mitochondrial genome of B. pectinirostris will provide useful genetic information for future phylogenetic and taxonomic classification of B. pectinirostris.

Similar but Different: Differences Hitherto Overlooked between Boleophthalmus pectinirostris and B. boddarti (Teleostei: Oxudercinae) in Indonesian Waters

During 2006-2017, we made a series of observations of Boleophthalmus pectinirostris on the east coast of South Sumatra province and Bangka island, Sumatra, Indonesia. This species is very similar to B. boddarti, in size, external morphological characters and behaviour. In this paper, we discuss the external morphological differences between B. pectinirostris and B. boddarti. Records of B. pectinirostris in southern Sumatra increase the known distribution range of this species 800 km southwards, indicating that this species was previously confused with B. boddarti in Indonesian waters.

Fasting affects the intestine and bacterial flora in mudskippers (Boleophthalmus pectinirostris) in semiaquatic and underwater conditions

Fasting alters the structure of the intestine and its bacterial diversity, affecting the metabolism of three nutrients (sugar, fat, and protein). However, very little information is available on the effects of fasting on fish intestinal structure, bacterial flora, nutrient metabolism, and inflammation. In this study, fasting modified the intestinal bacterial floral compositions and diversity in mudskippers (Boleophthalmus pectinirostris), and altered the dominant flora in both semiaquatic (SA) and underwater (UW) conditions. The length of the intestinal villi and the thickness of the intestinal mucosa/submucosa decreased significantly during fasting. The intestinal mucosa/submucosa thickness of the SA group was obviously thinner than that in the UW group. The constitutive expression of glucose, long-chain fatty acid, and amino acid transporter mRNA was detected by real-time quantitative PCR. Expression was dramatically increased in the intestine after fasting. Interestingly, the expression of most of the transporters mRNA associated with energy metabolism was higher in the SA group than that in the UW group. Fasting also induced a pro-inflammatory response in the early and middle stages and an anti-inflammatory response in the middle and late stages. The fish intestine is more likely to present pro-inflammatory symptoms in SA and anti-inflammatory symptoms in UW. The results demonstrated that fasting plays a crucial role in the modulation of structure and function in the intestine and bacterial flora. The function of the fish intestine recovers more quickly after fasting in SA than that in UW.

Exogenous inositol ameliorates the effects of acute ammonia toxicity on intestinal oxidative status, immune response, apoptosis, and tight junction barriers of great blue-spotted mudskippers (Boleophthalmus pectinirostris)

Ammonia toxicity can disrupt the intestinal health of aquatic animals. It is important to find substances that alleviate these adverse effects. The present study explored the possible protective role of myo-inositol (MI) in ammonia-induced toxicity in the fish intestine. Great blue-spotted mudskippers (Boleophthalmus pectinirostris) accumulated in artificial seawater (15‰ salinity, n = 600) were randomly selected and intraperitoneally injected with NaCl (0.68%) or MI (2.5 mg/g fish in 0.68% NaCl) then exposed to artificial seawater alone (NaCl and MI group) or seawater containing 57.025 mmol/L ammonium chloride (NH3 and NH3 + MI group). After a 24-h experiment, it showed that ammonia exposure down-regulated the mRNA expression levels of intestinal barrier function proteins (Zo-1, Ocln, Cldn-5, Cldn-12, and Cldn-15) and anti-inflammatory cytokines (Tgf-β and Il-10) while the acute ammonia stress up-regulated the apoptosis genes (p53, Bax, Caspase-3, and Caspase-9) and pro-inflammatory cytokines (Tnf-α and Il-1β). Furthermore, ammonia challenge also induced oxidative stress, as the malondialdehyde and the protein carbonyl contents were increased. In addition, ammonia stress down-regulated the antioxidant enzymes (Cu/Zn-Sod, Cat, Gpx, and Gst) activities as well as their gene transcription levels. The administration of the exogenous myo-inositol greatly ameliorated the ammonia-induced changes in redox capacity, immune response, apoptosis, inflammation, and tight junction barrier function to levels similar to those of the NaCl group. Furthermore, fish injected with MI alone showed no significant changes compared with the NaCl group. Taken together, pretreatment with myo-inositol had no obvious side-effects and effectively protected the mudskippers' intestine from the toxicity caused by acute ammonia stress.
